TF 1: High-pressure water jet cutting of native tissue slices of human lung

Fabrication and culture of high-precision native tissue slices produced by high-pressure water jet cutting for long-term intravital microscopy based analysis of innate immune mechanisms of human lung

 

Transfer project TF 1 aims to establish spectral confocal microscopy-based intravital microscopy of human alveolar tissue. Therefore, an apparatus for high-pressure water jet cutting of agarose-free peripheral human lung tissue will be established. Furthermore, a perfused chamber for intravital microscopy allowing for undisturbed subcellular imaging will be constructed and tested. In concurrence these innovations will considerably expand the analytical options of the SFB-TR84 to explore infectious processes and mechanisms in the human alveolar compartment. The combination of atraumatic tissue preparation via high-pressure water jet cutting with a specialized tissue culture chamber optimized for intravital imaging furthermore innovate the general field of ex vivo tissue culture and imaging as well as gives an outlook towards further human disease models.
